There are two types of column chromatography, according to the way how the solvent flows down the column. Gravity column chromatography, also known as percolation, is the process of allowing the solvent to flow down the column by gravity. The “state of the art” technique is called flash chromatography if the solvent is pushed down the column by positive air pressure. Currently, research labs for organic chemistry employ this method.
The Adsorbent
For column chromatography, organic chemists frequently utilize silica gel (SiO2) and alumina (Al2O3) as adsorbents. These adsorbents come in various mesh sizes, which are identified by numbers on the bottle label; for example “silica gel 60” and “silica gel 230-400.” Specifically, this number relates to the number of holes in the mesh or sieve that the raw silica particle combination passes through during the production process.
The solvent flows differently through the column depending on the adsorbent particle size. For flash chromatography, smaller particles with higher mesh values are applied; for gravity chromatography, larger particles with lower mesh values are utilized. For gravity columns, 70–230 silica gels are applied, whereas flash columns use 230–400 mesh.
